Single-use systems: the plastics that replaced stainless steel

Modern biologics plants are built around disposable plastic assemblies rather than fixed stainless steel. It removes cleaning and validation between batches and makes facilities flexible — and it converts a capital asset into a consumable supply chain that can run out.

The argument is compelling. The consequence is a permanent dependency. The bag film is a multi-layer polymer qualified for the process, and a change of film or supplier requires evidence that nothing leaches into the product and that the product is unchanged. During the last demand surge this became the visible constraint: facilities existed, and the disposables to run them did not.

Anything that can migrate from plastic into a drug product must be identified and shown to be safe at the levels present. That work is done per film, per process and per product, and it is the reason a manufacturer cannot substitute an apparently identical bag from another supplier when supply tightens.

Why the shortage was so hard to fix

Film production is specialised, qualification takes months to years, and every user had qualified a specific supplier. Adding capacity meant new extrusion lines and then a queue of customer qualification programmes. It is the same shape as a semiconductor materials shortage, for the same reason: the constraint is qualification, not manufacturing.

Where stainless steel remains

Very large scale still favours fixed vessels, because single-use bags have practical size limits and the consumable cost per batch grows with volume. High-volume established products therefore stay in stainless steel, while newer products and contract facilities are predominantly single-use.

Why not just buy bags from another supplier during a shortage?
Because the specific film is qualified into the process with evidence that nothing harmful migrates into the product. A different film requires that work to be redone and, often, a regulatory filing. Manufacturers were physically able to buy alternatives and not able to use them.
Is single-use always better?
For multi-product and mid-scale facilities, generally yes — no cleaning validation, faster changeover, lower capital. At very large scale for a single established product the consumable cost per batch and practical bag size limits favour stainless steel, which is why both coexist.
